More...This study aimed to identify key gut microbial species and metabolites associated with the chemopreventive effects of JWJGC against RA, utilizing 16S rRNA, metagenomics sequencing, metabolomics analysis, antibiotic treatment, and fecal microbiota transplantation (FMT) in a collagen-induced arthritis (CIA) rat model. Additionally, transcription analysis of colon and synovium, as well as intestinal barrier permeability testing, were performed to investigate how JWJGC regulates gut microbiota to reduce RA severity. Cell Counting Kit (CCK)-8 assay was applied to detect the effect of JWJGC on the viability of Caco2 cells. Moreover, lipopolysaccharides (LPS)-induced Caco-2 cell monolayer model of intestinal barrier was established in vitro, with administration of either blank serum or JWJGC, to examine JWJGC's effects on transepithelial electrical resistance (TEER), apparent permeability coefficient (Papp), inflammatory cytokines, and tight junction proteins (TJs).
