More...Despite significant advances in medical technology and treatment strategies in recent years, the prognosis for patients with esophageal cancer remains unsatisfactory. Moreover, the pathogenesis and therapeutic targets of esophageal cancer remain unclear. APE1 plays a role in chemotherapy resistance and radiotherapy development due to its abnormal expression and subcellular localization in various cancers. Hence, APE1 is an emerging therapeutic target for treating multiple cancers. Prior research revealed that in esophageal cancer, elevated APE1 can lead to homologous recombination and cell cycle dysregulation, resulting in genome instability and tumorigenesis. However, so far, no relevant reports have elucidated the impact of APE1 knockdown on the development and function of esophageal cancer cells. In this study, sh-APE1 and sh-NC esophageal cancer cell lines are constructed. RNA-seq technology is used to sequence the transcriptomes of the two cell groups to obtain comprehensive gene expression information. Then, bioinformatics analysis was performed on the differentially expressed genes, further confirm the results of RNA-Seq analysis using qRT-PCR and Western blotting analysis. The findings of this study demonstrate the differential gene expression and potential functions of shAPE1 in esophageal cancer cells, providing new understanding and targets for the pathogenesis and treatment of esophageal cancer.
