After 60 hours of infection host cells were removed by filtration and parasite spores were incubated for a further 48 hr. cDNA was PCR amplified using a trans-spliced leader primer and a reverse primer from the polyT reverse transcriptase primer. Total RNA was used as a template and sequences from both host and parasite are present in the library. These are putative parasite sequences based on AT >= 60%.
