K562 Gene Expression Control vs 1R-Chl treatments

(Submitter supplied) To determine whether the polyamide-Chl conjugate 1R-Chl would cause similar changes in global gene expression in K562 cells, affymetrix gene chip analysis was performed using 1R-Chl. Through class comparison analysis, 1R-Chl affected the levels of transcription and genes of interest were determined. Keywords: Treatment response

Targeted suppression of metastasis controlling transcription factor SOX2 in various cancers using a sequence-specific designer Pyrrole-Imidazole Polyamide

(Submitter supplied) SOX2 is a well-known transcription factor that plays a central role in maintaiing self renewal, pluripotency of embryonic stem cells, and cell fate determination during embryonic development. N-Methylpyrrole-N-methylimidazole polyamides (PIPs) are chemical compounds that can selectively recognize DNA sequence and achieve operative control over gene expression. In this study, the antitumor activity of a PIP targeting SOX2 binding sequence (SOX2i), which was recently shown to induce the targeted differentiation of stem cells into cardiomyocytes, was evaluated.

Direct Targeting of MYCN Gene Amplification by CRISPR/Cas9 in Neuroblastoma Cells (CHP-134)

(Submitter supplied) Amplification of MYCN plays a pivotal role in multiple types of tumors and correlates with poor prognosis in high-risk neuroblastoma. Despite recent advances in the treatment of neuroblastoma, no approaches directly target the master oncogene MYCN. Difficulties in targeting the MYCN protein have inspired us to develop a new gene level inhibitory strategy using a sequence-specific gene regulator. Here we generated a MYCN-targeting pyrrole-imidazole (PI) polyamide, MYCN-A3, which directly binds to and alkylates DNA at homing motifs within MYCN transcript. more...

Gene expression signatures for combined ATO and CDDP treatment (Ato-C) in CML cell lines

(Submitter supplied) To investigate the effect of Ato-C on gene expression, we have employed whole genome microarray expression profiling as a discovery platform to identify genes with the potential to suppress the cell proliferation of Ph+ CML cell lines. K562, KU-812 and MEG-A2 cells were treated with ATO, CDDP or Ato-C for 24 h in vitro.

Genomic expression analysis of K562 cells expressing shRNA targeting lncRNA-IIRX and control cells

(Submitter supplied) LncRNA-IIRX plays critical role in Bcr-Abl-induced tumorigenesis. To discover its mechanisms underlying cellular transformation by Bcr-Abl oncogene, genome-wide mRNA expression was measured by RNA-seq in K562 cells expressing shRNA targeting lncRNA-IIRX and control cells. We identified many genes with differential expression in K562 cells after knocking down lncRNA-IIRX.

Identification of a novel lncRNA family that is required for efficient cellular transformation by Bcr-Abl oncogene

(Submitter supplied) Aberrantly expressed long noncoding RNAs (lncRNAs) have been described in diverse human diseases and cancer development. Chronic myeloid leukemia (CML) is a hematological malignancy induced by Bcr-Abl hybrid gene. Owing to the development of tyrosine kinase inhibitors (TKIs), especially the first-generation Imatinib, over 90% of CML patients can be cured in recent years. Here we attempt to identify Imatinib-inducible lncRNAs associated with CML by analyzing lncRNA expression profiles in K562 cells after Imatinib or control treatment. more...
