Radiation-Induced Changes in Gene Expression Involve Recruitment of Existing Messenger RNAs to and from Polysomes

(Submitter supplied) Although ionizing radiation has been shown to influence gene transcription, little is known about the effects of radiation on gene translational efficiency. To obtain a genome-wide perspective of the effects of radiation on gene translation, microarray analysis was done on polysome-bound RNA isolated from irradiated human brain tumor cells; to allow for a comparison with the effects of radiation on transcription, microarray analysis was also done using total RNA. more...

Colon cancer progression-polysomal difference

(Submitter supplied) Two colon cancer cell lines are under study. SW480 and SW620. The first one is derived from primary cancer, SW620 are from lymphnode metastatic sites. they both comes from the sampe patient. Polisomal RNA fractions from the two isogenic colon cancer cells lines was purified by sucrose gradient and hybridized on affymetrix hgu133a chips. this study is complementary to the series GSE1323 were total RNA was used instead. more...

Isogenic primary tumor/metastasis comparison

(Submitter supplied) SW480 and SW620 are colon cancer cells lines derived from a primary tumor and a corresponding metastasis from the same individual. The numbers indicate the three indipendent replicate RNA samples processed. Three different software packages were used in parallel for signal calculation: Affymetrix microarray suite 5.0, DNA-Chip analyzer, and Robust multi-array analyses. Keywords = metastasis Keywords = colorectal cancer Keywords = tumor progression Keywords: repeat sample

Colorectal cancer progression: polysomal mRNA profiles

Comparison of polysomal RNA from isogenic cell lines established from a colorectal cancer (CRC) patient. SW480 and SW620 cells derived from the primary tumor and a lymph node metastasis, respectively. The cell lines constitute a cellular model of CRC transition from invasive carcinoma to metastasis.

Hep3B time-course

(Submitter supplied) A time-course (T0, 15, 30 min, 1, 3, 6 or 16 hours) of mRNA abundance changes was studied in the human Hep3B hepatoma cell line challenged with a pro-inflammatory cytokine-enriched medium (CM,conditionned medium) vs control medium (NCM,non conditionned medium). Paired cultures prepared from a same batch of trypsinized cells were challenged with CM vs NCM for a given length of time and this was repeated in three independent experiments (S1,S2,S3). more...

Expression data from non-infected and Salmonella Choleraesuis infected mesenteric lymph nodes

(Submitter supplied) To understand the host transcriptional response to S. enterica serovar Choleraesuis (S. Choleraesuis), the first generation Affymetrix porcine GeneChip® was used to identify differentially expressed genes in the mesenteric lymph nodes responding to infection at acute (8 hours (h), 24h, 48h post-inoculation (pi)) and chronic stages (21 days (d) pi) The objectives of this study were to identify and examine the stereotypical gene expression response within the host mesenteric lymph nodes to S. more...

Gene expression in the liver following exposition to 1Gy ionizing radiation

(Submitter supplied) Although major differences in the response of tissues were documented in the literature, the molecular mechanisms underlying tissue radiosensitivity are still fragmentary and need to be study more thoroughly. To this end, a genome wide analysis strategy was used to investigate changes in gene expression in the liver following exposition to ionizing radiation. This follows the tissue survey that was previously initiated with other organs and tends to obtain a comprehensive picture of the molecular events underlying tissue radio sensitivity. more...

Integrated analyses of early responses to radiation in glioblastoma identify new alterations in RNA processing and candidate targets to improve treatment outcomes

(Submitter supplied) High-dose radiation is the main component of glioblastoma therapy. Unfortunately, radio-resistance is a common problem and a major contributor to tumor relapse. Understanding the molecular mechanisms driving response to radiation is critical for identifying regulatory routes that could be targeted to improve treatment response. We conducted an integrated analysis in the U251 and U343 glioblastoma cell lines to map early alterations in the expression of genes at three levels: transcription, splicing, and translation in response to ionizing radiation.
