Microarray analysis of differentiation of human glioblastoma stem cells

(Submitter supplied) Glioblastoma multiforme is one of the most devastating cancers and presents unique challenges to therapy due to its aggressive behaviour. Cancer stem cells have been described to be the only cell population with tumorogenic capacity in glioblastoma. Therefore, effective therapeutic strategies targeting these cells may be beneficial. We have established different cultures of glioblastoma stem cells (GSCs) derived from surgical specimens and found that, after induction of differentiation, NFκB was activated, which allows intermediate tumor precursor cells to remain cycling. more...

Expression data of glioblastoma stem-like (GS) cell lines, conventional glioma cell lines and primary tumors

(Submitter supplied) We compared a large panel of human glioblastoma stem-like (GS) cell lines, corresponding primary tumors and conventional glioma cell lines to identify cell lines that preserve the transcriptome of human glioblastomas most closely, thereby allowing identification of shared therapeutic targets. We used Affymetrix HG-U133 Plus 2.0 microarrays to compare human glioblastoma stem-like (GS) cell lines, corresponding primary tumors and conventional glioma cell lines.

Expression analyses of glioblastoma derived neurosphere cultures

(Submitter supplied) Recent studies demonstrated that tumor cells with stem cell-like properties can be cultured from human glioblastomas by using conditions that select for the expansion of neural stem cells. We established glioblastoma stem-like (GS-) cell cultures from 9 different glioblastomas, 8 of which generated stably expandable cell lines. Analyzing GS-cell cultures, we discovered two clearly discernable phenotypes. more...

Stable XIAP knockdown in HCT116 colon cancer cells

(Submitter supplied) X-linked inhibitor of apoptosis (XIAP) is the most potent endogenous caspase inhibitor preventing cell death via caspase-9, -7 and -3 (initiator and executioner caspase pathways). Using short hairpin RNA (shRNA) against XIAP, stably expressed in a parent HCT116 human colon cancer cell line, a series of clones have been developed. XIAP mRNA levels were established by RT-PCR, the four X (XIAP knockdown) clonal cell lines show 82-93% reduction in XIAP mRNA when compared to the four L (luciferase control) cell lines. more...

Gene expression profiles of progestin-induced canine mammary hyperplasia and spontaneous mammary tumors

(Submitter supplied) Spontaneous mammary tumours are the most prevalent type of neoplasm in women as well as in female dogs. Even though ovarian hormones, mainly estrogens and progestins, are known to play a key role in mammary tumorigenesis, conflicting reports have been obtained from in vivo and in vitro studies concerning their role, especially of progestins, in mammary tumorigenesis. Prolonged exposure to high levels of progestins during unusually long luteal phase of oestrous cycle is suspected to be the key event in canine mammary tumorigenesis. more...

Delayed cell cycling in DS Ts1Cje neural precursor cells results in gene expression dysregulation

(Submitter supplied) Down syndrome neurophenotypes are characterized by mental retardation and a decreased brain volume. In order to identify whether deficits in proliferation, differentiation or survival could be responsible for this phenotype, neural precursor cells (NPCs) were isolated from the developing E14 neocortex of Down syndrome partial trisomy Ts1Cje mice and euploid (WT) littermates. Proliferation, cell differentiation and cell death assays revealed that Ts1Cje NPCs proliferated at a slower rate, due to a longer cell cycle and that a greater number of cells were positive for glial fibrillary acidic protein. more...

Novel Lipogenic Enzyme ELOVL7 Involves in Prostate Cancer Growth through Saturated Fatty-acid and Steroid Metabolism

(Submitter supplied) We identified a novel lipogenic gene, ELOVL7, as an over-expressed gene in prostate cancer (PC) cells and it is involved in elongation of long-chain fatty acids and can play some important roles in prostate carcinogenesis. To investigate into downstream pathways of ELOVL7, we performed the gene expression analysis by the genome-wide cDNA microarrays. The genome–wide gene expression patterns were compared between PC cells transfected with siELOVL7 and these with siCONTROL and between the ELOVL7-overexpressing in-vivo tumors in mice bread with high fat diet (HFD) and those with normal diet (ND). more...

Profiling of spatio-temporally regulated transcripts during cerebral corticogenesis in the mouse

(Submitter supplied) The cerebral cortex plays an important role in cognitive function and specialized perception in mammals and its development requires highly specific spatio-temporal control of gene expression. The study identified stage- and region-specific markers throughout cerebral corticogenesis at various important stages of cerebral cortex development; embryonic day (E) 15.5, E17.5, postnatal day (P) 1.5 and 4-6 months old.
