Molecular Changes of Lung Malignancy in HIV Infection

(Submitter supplied) To evaluate the molecular changes of lung malignancy in HIV infection, we analyzed the differential gene expression profiles and screened the early detection biomarkers of HIV-associated lung cancer using Affymetrix arrays. RNA was for transcriptomic profiles analysis in the HIV lung cancer tumor tissue samples from 3 patients with AC and 1 patient with squamous cell carcinoma (SCC) including 3 pairs of tumor/adjacent normal tissue paraffin specimens and 1 pair of tumor/adjacent normal fresh tissue samples.

Affymetrix SNP array data for NSCLC samples

(Submitter supplied) Non-small cell lung cancer (NSCLC) presents a notoriously genomically unstable cancer type, with numerous large scale and focal genomic aberrations resulting in gene copy number variations across the whole genome. The relations of these gene copy number changes to subsequent mRNA levels are only fragmentarily understood. The aim of this study was an integrated analysis of gene copy number changes and corresponding gene expression in a large clinically annotated NSCLC patient cohort. more...

Gene Copy Number Aberrations are Associated with Survival in Histological Subgroups of Non-Small Cell Lung Cancer (copy number data)

(Submitter supplied) Hypothesis: Non-small cell lung cancer (NSCLC) is characterized by a multitude of genetic aberrations with unknown clinical impact. In this study, we aimed to identify gene copy number changes that correlate with clinical outcome in NSCLC. To maximize the chance to identify clinically relevant events, we applied a strategy involving two prognostically extreme patient groups. Results: Genetic aberrations were strongly associated with tumor histology. more...

Cancer-Associated Fibroblasts Support Lung Cancer Stemness through Paracrine IGF-II/IGF1R/Nanog Signaling

(Submitter supplied) The CLS1/CAF co-culture maintained the cancer stemness. This cancer stemness was lost when the CAF feeder cells were removed during passaging. To investigate the specific signaling pathways and markers of lung CSCs, we analyzed the gene expression profile of the CLS1 sphere after CLS1/CAF co-culture and compared this profile with that obtained for CLS1 cells cultured without feeder cells through different passages (CLS1 p3, p6, and p14) and CLF1

High risk HPV-positive lung carcinomas: molecular evidence for a pattern of pulmonary metastasis

(Submitter supplied) BACKGROUND: Infection with high-risk types of human papillomavirus (hrHPV) is associated with cervical, anogenital and oropharyngeal cancers. A role for hrHPV in lung cancer has been proposed, although previous reports on the contribution of hrHPV infection to lung cancer have provided contradictory results. The current study evaluated hrHPV prevalence in lung tumor specimens of different histological subtypes in a Western study population using a validated test algorithm for hrHPV detection in formalin-fixed paraffin-embedded (FFPE) tumor specimen (i.e., GP5+6+-PCR and p16INK4A immunohistochemistry). more...

Claudin-1 is an Invasion and Metastasis Suppressor of Lung Adenocarcinoma

(Submitter supplied) Background: Claudin-1(CLDN1), one of the key components of tight junction proteins, was found to be down-regulated in human lung adenocarcinomas. This study we investigated the clinical significance of CLDN1 expression in lung adenocarcinoma patients and its role in cancer progression. Method: CLDN1 mRNA expression was measured in tumor specimens from 51 patients with lung adenocarcinoma. CLDN1 protein expression was also examined by immunohistochemistry on specimens from an independent cohort of 67 lung adenocarcinoma patients. more...

Claudin-1 overexpression effect on lung adenocarcinoma cell line

Analysis of lung adenocarcinoma CL1-5 cells overexpressing Claudin-1 (CLDN1), a component of tight junction complexes. Low CLDN1 expression in lung adenocarcinomas is associated with shorter overall survival. Results provide insight into the role of CLDN1 in the progression of lung adenocarcinoma.

Identification of miR-145 or miR-133a target genes in squamous cell carcinoma

(Submitter supplied) We recently identified a subset of down-regulated miRNAs such as miR-145 and miR-133a in squamous cell carcinoma. Cell growth inhibitions occurred in miR-145 and miR-133a transfectants compared with the controls, suggesting that both miRNAs function as tumor suppressors. The aims of our expression studies were identification of these miRNAs target genes.

Comprehensive RNA-Seq Reveals Molecular Changes in Kidney Malignancy Among People Living with HIV

(Submitter supplied) To heighten the awareness of kidney malignancy in patients with HIV infection to facilitate the early diagnosis of kidney cancer, the differentially expressed mRNAs were analyzed in this malignant tumor using RNA sequencing (RNA-seq). We identified 2962 protein-coding transcripts in HIV-associated kidney cancer. KISS1R, CAIX, and NPTX2 mRNA expression levels were specifically increased in HIV-associated kidney cancer, and UMOD and TMEM213 mRNA were decreased in most cases based on real-time PCR analyses. more...
