Analysis of MACC1 dependent gene expression regulation in CRC cells

(Submitter supplied) We employed whole genome expression profiling to identify differential gene expression in the colorectal cancer (CRC) cell line SW480 (ATCC - CCL-228), dependent on the expression level of MACC1 (Metastasis Associated in Colon Cancer 1). SW480 cells with endogenously low expression levels of MACC1 were transfected either with CMV-promoter driven MACC1-cDNA or the empty vector, and selected for stable expression.

Predicting Impact of Myeloid Cells on T Cells Effector Function from Spatial Organization of TiME in CRC

(Submitter supplied) Recent developments in digital computational pathology support not only classical cell density based tumor characterization, but also a more comprehensive analysis of the spatial cell organization in the tumor immune microenvironment (TiME). Leveraging that methodology in the current study, we tried to address the question of how the distribution of myeloid cells in TiME of primary colorectal cancer (CRC) affects the function and location of cytotoxic T cells. more...

Revealing and harnessing CD39 for the treatment of colorectal cancer and liver metastases by engineered T cells [NanoString]

(Submitter supplied) Colorectal tumors are often densely infiltrated by immune cells that have a role in surveillance and modulation of tumor progression but are burdened by exhaustion mechanisms fueled by the tumor microenvironment, which must be counteracted to achieve control. Here, we deployed a multidimensional approach to unravel the T cell functional landscape in tumor and peritumoral tissues from primary colorectal cancers and liver metastases. more...

Clinical significance of UNC5B Expression in Colorectal Cancer

(Submitter supplied) Purpose and Experimental Design: The purpose of this study is to find a methylation-related gene that could become a biomarker or therapeutic target in colorectal carcinoma (CRC). We screened candidate genes suspected to be silenced by DNA methylation using oligonucleotide microarray analysis. To investigate the clinical significance of one candidate gene (UNC5B), we analyzed the correlation between mRNA expression and clinicopathological features using clinical tissue samples. more...

STING suppresses the reactivation of dormant metastasis

(Submitter supplied) Metastatic relapse frequently develops from disseminated cancer cells that remain dormant in distant organs after the apparently successful treatment of a primary tumor. Disseminated cancer cells fluctuate between immune evasive quiescent and cell cycle reentry states, which exposes them to elimination by the immune system. Little is known about the molecules that determine immune-mediated clearing of awakened metastatic cells and how this process could be therapeutically activated to eliminate residual disseminated disease in patients. more...
