COP1 depletion effect on hepatocellular carcinoma cell lines

Analysis of Huh7, HepG2, and Hep3B hepatocellular carcinoma (HCC) cells depleted for the ubiquitin modulator COP1. COP1 regulates p53 activity by ubiquitination. p53 is wild type in HepG2, mutated in Huh7, and lacking in Hep3B. Results provide insight into the role of COP1 in HCC pathogenesis.

We have previously demonstrated that constitutively photomorphogenic 1 (COP1), which regulates p53 activity by ubiquitination, is frequently overexpressed in human HCC. Molecular targeting of CSN5 in human hepatocellular carcinoma: a mechanism of therapeutic response

(Submitter supplied) Development of targeted therapy for hepatocellular carcinoma (HCC) remains a major challenge. We have recently identified an elevated expression of the fifth subunit of COP9 signalosome (CSN5) in early HCC as compared to dysplastic stage. In the present study, we explored the possibility of CSN5 being a potential therapeutic target for HCC. Transcriptomic profiling of Cop1-deficient embryos

(Submitter supplied) In order to assess the physiological role of Cop1 in vivo we generated mice that do no longer express the protein. Cop1KO mice die at around E10.5 of embryonic development. In order to gain insights into the molecular mechanisms that cause the embryonic death we compared the genome-wide gene expression profile of E9.5 wild-tytpe and Cop1-null embryos. The data do not support a role for Cop1 in the regulation of the p53 pathway in vivo and highlight a role for Cop1 in cardiovascular development and/or angiogenesis. more...
