Running effect on hippocampal subgranular zone: time course

Analysis of LCM samples of the subgranular zone (SGZ) of the hippocampal dentate gyrus isolated from adults subjected to running for up to 30 days. The SGZ is a major site of neurogenesis in adult brain. Running enhances SGZ proliferation. Results provide insight into the SGZ molecular environment.

Molecular Signatures of Neurogenesis in the Hippocampal Subgranular Zone of Rodents

(Submitter supplied) We sought to find molecular signatures of the SGZ cell types, and to characterize the molecular pathways and transcription factor cascades that define the neurogenic niche. We used laser capture microdissection and DNA microarrays to profile gene expression in the inner (SGZ) and outer portions of the dentate gyrus (DG). Since the vast majority of the cells in the DG are mature granule cells, we compared the expression of the inner and outer portions to reveal molecular markers for the less numerous populations of the SGZ. more...

Profiling of spatio-temporally regulated transcripts during cerebral corticogenesis in the mouse

(Submitter supplied) The cerebral cortex plays an important role in cognitive function and specialized perception in mammals and its development requires highly specific spatio-temporal control of gene expression. The study identified stage- and region-specific markers throughout cerebral corticogenesis at various important stages of cerebral cortex development; embryonic day (E) 15.5, E17.5, postnatal day (P) 1.5 and 4-6 months old.

Sox11 loss-of-function mutant cortex at E17.5

(Submitter supplied) Expression profiling of cortex from embryonic day (E) 17.5 telencephalon of Sox11(+/+) and Sox11(-/-) mouse embryos. Sox11 is implicated in regulating proliferation, neuronal migration and differentiation. Single cell sequencing of the hippocampal niche

(Submitter supplied) Adult neurogenesis in the murine dentate gyrus occurs in a specialized microenvironment that sustains the generation of neurons during life. To fully understand adult neurogenesis, it is essential to determine the neural stem cell (NSC) and progenitor developmental stages, their molecular determinants, and the niche cellular and molecular composition. We report on a single cell RNA sequencing study of the hippocampal niche, performed by isolating all the non-neuronal cell populations. more...

Identification of SoxC-regulated genes during neurogenesis in the developing spinal cord [chicken]

(Submitter supplied) The HMG-domain containing SoxC transcription factors Sox4 and Sox11 are expressed in the vertebrate central nervous system in neuronal precursors and neuroblasts. They are required during early stages of neurogenesis. Here we perform micorarray analysis to identify genes that are downstream of these SoxC proteins during spinal cord neurogenesis in chicken. The identified genes represent potential direct target genes of Sox4 and Sox11.

Regulation of gene expression in the postnatally developing monkey hippocampal formation

(Submitter supplied) The hippocampus is part of a brain network essential for memory function. Paradoxically, the hippocampus is also the brain structure that is most sensitive to hypoxic-ischemic episodes. Here we show that the expression of genes associated with glycolysis and glutamate metabolism in astrocytes and the coverage of excitatory synapses by astrocytic processes undergo significant decreases in the CA1 field of the monkey hippocampus during postnatal development. more...

Single-cell transcriptomic analysis of craniogenesis in control and SOXC-mutant mice

(Submitter supplied) SOXC genes (i.e., Sox4, Sox11 and Sox12) are important members of the SOX transcription factor family. They are involved in several developmental processes, including skeleton formation. However, their role in intramembranous ossification and, particularly, in skull development has not beed investigated yet. Here, we employed single-cell RNA-seq technology to better understand the roles of the SOXC genes in craniogenesis and how their inactivation could affect this process. more...

Transcriptomic Taxonomy and Neurogenic Trajectories of Adult Human, Macaque and Pig Hippocampal and Entorhinal Cells

(Submitter supplied) The hippocampal-entorhinal system supports cognitive functions, has lifelong neurogenic capabilities in many species, and is selectively vulnerable to Alzheimer’s disease. To investigate neurogenic potential and cellular diversity, we profiled single-nucleus transcriptomes in five hippocampal-entorhinal subregions in human, macaque, and pig. Integrated cross-species analysis revealed robust transcriptomic and histologic signatures of neurogenesis in adult mouse, pig and macaque, but not humans. more...
