Polybrominated diphenyl ether effect on the liver

Analysis of livers collected from postnatal day (PND) 22 and 13-week-old Wistar/Han rats exposed in utero to polybrominated diphenyl ether (PBDE) and directly dosed with PBDE. PBDEs are flame retardants. Results provide insight into the molecular basis of PBDE toxicities.

Expression data from PBDE treated rats at PND22 and Week 13

(Submitter supplied) Liver gene transcripts patterns were used to characterize toxicity from exposure to polybrominated diphenyl ethers (PBDEs), flame retardant components. In this study, Wistar Han dams were exposed by gavage to the PBDE mixture (DE71) starting at gestation day 6 (GD 6) and continuing to weaning on postnatal day 21 (PND 21). Offspring from the dams began PBDE direct dosing on PND 12 and were dosed daily through PND 21. more...

Expression data from DE-71 treated rats at PND4

(Submitter supplied) To identify liver transcripts differentially expressed between control samples and animals exposed to DE-71, we collected RNA from male pups at postnatal day 4 (PND4) after the Wistar Han dams were exposed to 0, 0.1, 15, or 50 mg/kg DE-71. These samples were interrogated with the Affymetrix Rat Genome 230 2.0 GeneChip array A total of 0, 15, and 162 gene transcripts were differentially expressed between control samples and 0.1, 15, and 50 mg/kg DE-71, respectively (using a false discovery rate (FDR) threshold of 0.05). more...

Liver of rat offsprings exposed perinatally to low doses of 2,2',4,4'-tetrabromodiphenyl ether

(Submitter supplied) Background: Polybrominated diphenyl ethers are a group of flame-retardant chemicals appearing increasingly in the environment. Their health effects and mechanisms of toxicity are poorly understood. Objectives: We screened for the sensitive effects and mechanisms of toxicity of 2,2 ,4,4 -tetra-bromodiphenyl ether (BDE-47) by analyzing the gene expression profile in rats exposed to doses comparable to human exposure. Methods: Wistar dams were exposed to vehicle or BDE-47 (0.002 and 0.2 mg/kg body weight) every fifth day from gestation day 15 to postnatal day 20 by injections to caudal vein. more...

Liver response to perinatal exposure to 2,2',4,4'-tetrabromodiphenyl ether: dose response

Analysis of livers of animals exposed to low doses of 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) from gestational day 15 to post-natal day 20. Polybrominated diphenyl ethers are flame-retardant chemicals used in consumer products. Results provide insight into the mechanisms of toxicity of BDE-47.

Novel Interactions between Gut Microbiome and Host Drug-processing Genes Modify the Hepatic Metabolism of the Environmental Chemicals PBDEs

(Submitter supplied) Purpose: Next-generation sequencing (NGS) has been utilized for systems-based analysis of all liver samples. The goals of this study were to compare the hepatic transcriptome and PBDE metabolism between conventional (CV) and germ-free (GF) mice. Methods: Livers from vehicle (corn oil), BDE-47, or BDE-99 treated adult male CV and GF mice were used for RNA-Seq (biological replicates: n=3 for CV corn oil, n=4 for CV BDE-47, n=2 for CV BDE-99, n=3 for GF corn oil, n=3 for GF BDE-47, and n=3 for BDE-99) using a HiSeq 2000 sequencer. more...

Maternal Western-style high fat diet induces sex-specific physiological and molecular changes in two-week-old mouse offspring

(Submitter supplied) Maternal diet is associated with the development of metabolism-related and other non-communicable diseases in offspring. Underlying mechanisms, functional profiles, and molecular markers are only starting to be revealed. Here, we explored the physiological and molecular impact of maternal Western-style diet on the liver of male and female offspring. C57BL/6 dams were exposed to either a low fat/low cholesterol diet (LFD) or a Western-style high fat/high cholesterol diet (WSD) for six weeks before mating, as well as during gestation and lactation. more...
