Expression Profiles of Rats Treated with Clofibric Acid: Comparison of Whole and Laser Capture Microdissected Liver

(Submitter supplied) This is an evaluation of the impact of the successive steps of LCM procedure on gene expression profiling by comparing profiles from LCM samples to those obtained with non-microdissected liver samples collected after a one month CLO treatment in the rat. Long-term Clofibric acid study

(Submitter supplied) F344 rats were divided into 4 groups: vehicle control, DEN, DEN+CLO and CLO. After one week of basal diet, rats belonging to the groups DEN and DEN+CLO underwent intraperitoneal injection of DEN (30mg/kg body weight) dissolved in NaCl 9‰. The two other groups were injected with NaCl 9‰ alone. At 30mg/kg, DEN is non-necrogenic thus only exhibiting initiating properties. Twelve days after injection, diet from rats belonging to the groups CLO and DEN+CLO was changed for diet containing 5000ppm CLO for up to 608 days. more...

Ciprofibrate stimulated gene expression in rats

(Submitter supplied) Samples and RNA preparation: The series is composed of 4 hybridizations for analysis of differentially expressed genes in the liver of ciprofibrate dosed vs. control rats. Rats (200-250 g) were given (by gastric intubation) tap water (control group, n=5) or ciprofibrate (50 mg/kg body weight, once daily for 60 days, n=4). Ciprofibrate (PPAR-alpha agonist) in the monkey liver

(Submitter supplied) Liver gene expression was examined in male cynomolgus monkeys treated with ciprofibrate (PPAR-alpha agonist) for 4 days at 400 mg/kg/day and treated for 15 days at 0, 3, 30, 150 or 400 mg/kg/day. The untreated control group were given only the vehicle (0.5% hydroxypropyl methylcellulose). PPARα agonist ciprofibrate effect on liver

Analysis of primate livers exposed to ciprofibrate at various doses for 4 or 15 days. Peroxisome proliferator-activated receptor-α (PPARα) agonists such as ciprofibrate cause hepatocarcinogenesis in rodents but not in primates. Results identify species differences in response to PPARα agonist.
