Anaplastic thyroid carcinomas: thyroid biopsies

Analysis of anaplastic thyroid carcinomas (ATC), fresh-frozen and fine-needle aspiration biopsies. ATC is a rare but highly lethal form of thyroid cancer. Results provide insight into the molecular alterations contributing to ATC development.

Gene expression profilling of anaplastic thyroid carcinomas (ATC) and normal thyroid tissues

(Submitter supplied) ATC are among the most lethal malignancies, for which there is no effective treatment. ATC global gene expression was profiled and compared to normal thyroid tissues, in order to elucidate the molecular alterations contributing to ATC development, and to identify novel therapeutic targets.

Genomic and Transcriptomic Hallmarks of Poorly-Differentiated and Anaplastic Thyroid Cancers

(Submitter supplied) BACKGROUND. Poorly-differentiated (PDTC) and anaplastic (ATC) thyroid cancers are rare and frequently lethal tumors, which so far have not been subjected to comprehensive genetic characterization. METHODS. We performed next generation sequencing of 341 cancer genes in 117 PDTCs and ATCs, and a transcriptomic analysis of a representative subset of 37 tumors. Results were analyzed in the context of The Cancer Genome Atlas (TCGA) study of papillary thyroid cancers (PTC). more...

Expression analysis of mouse thyroid tumors

(Submitter supplied) Anaplastic thyroid carcinoma (ATC) is the most aggressive form of thyroid cancer, and often derives from pre-existing well-differentiated tumors. We have engineered the first mouse model of ATC by combining in the mouse thyroid follicular cells two molecular hallmarks of human ATC: activation of PI3K (via Pten deletion) and inactivation of p53. By 9 months of age, over 75% of the compound mutant mice develop aggressive, undifferentiated thyroid tumors that evolve from pre-existing follicular hyperplasia and carcinoma. more...

Human thyroid adenomas, carcinomas, and normals

(Submitter supplied) Human samples of various thyroid carcinomas, adenomas, and normals, each from a different patient, had mRNA assays performed using Affymetrix HG_U133A arrays, with 22283 probe-sets. The 99 samples consisted of 4 normals, 10 follicular adenomas, 13 follicular carcinomas, 7 oncocytic adenomas, 8 oncocytic carcinomas, 51 papillary carcinomas (each typed as having classical, follicular or tall cell morphology), 4 anaplastic carcinomas, and 2 medullary carcinomas. more...

Gene expression profiling associated with the progression to poorly differentiated thyroid carcinomas

(Submitter supplied) Poorly differentiated thyroid carcinomas (PDTC) represent a heterogeneous, aggressive entity, presenting features that suggest a progression from well-differentiated carcinomas. To elucidate the mechanisms underlying such progression and identify novel therapeutical targets, we assessed the genome-wide expression in normal thyroid tissues, well-differentiated thyroid carcinomas and PDTC.

Gene expression of thyroid cancer cell lines

(Submitter supplied) Purpose: The primary goal of this study was to identify gene-expression profiles of anaplastic thyroid cancer and to identify some novel in-frame gene fusions that could result in translated protein products affecting the development of anaplastic thyroid cancer. Methods: RNAseq Data was processed with TCGA UNC V2 RNAseq protocol and different expressed genes were identify by using DESeq2, limma-voom, and edgeR. more...

“Reprogram enablement” as an assay for identifying pivotal early oncogenic pathways by their ability to allow neoplastic cells to reacquire a normal epiblast-emulating state: evidence from human thyroid cancer

(Submitter supplied) One approach to understanding how tissue-specific cancers emerge is to determine the requirements for “reprograming” such neoplastic cells back to their developmentally-normal primordial premalignant epiblast-like pluripotent state and then scrutinizing their spontaneous reconversion to a neoplasm, perhaps rendering salient the earliest pivotal oncogenic pathway(s) (before other aberrations accumulate in the adult tumor). more...

Global DNA methylation patterns in primary anaplastic thyroid cancer

(Submitter supplied) Anaplastic thyroid cancer (ATC) is one of the most malignant tumors, with a median survival of only a few months. The tumorigenic processes of this disease has not yet been completely unraveled. Here, we report an DNA methylation analysis of ten primary ATC. ATC displayed a global hypomethylation of the genome but with hypermethylation of promoters and CpG islands. This study shows that aberrant DNA methylation is common in ATC and likely contributes to tumorigenesis in this disease. more...
