C57BL/6J mice subject to TAC (transverse aortic constriction) were treated with BIX-01294 for 1 week via subcutaneous mini-osmotic pumps. The control groups included cardiomyocyte-enriched populations isolated from DMSO-treated mice after TAC and from sham surgery mice.

Extraction protocol |
RNA was extracted with Trizol reagent, followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyser.

The data exported from GenomeStudio V2011.1 were analysed with Lumi/Limma in R. Data preprocessing was performed using lumi package: - Background Correction: bgAdjust - Variance Stabilizing Transform method: vst - Normalization method: quantile The differentianly expressed genes were identified using limma package.
