Growth protocol |
Peripheral blood mononuclear cells first isolated by density centrifugation on Lymphoprep (Axis-Shield Diagnostics, Dundee, UK) underwent positive selection using anti-CD19 magnetic microbeads (Miltenyi Biotec, Bergisch, Gladback, Germany); median cell purity was 96.4% by flow cytometry. Total B lymphocyte RNA was immediately extracted using the AllPrep DNA/RNA Mini kit (Qiagen GmbH, Germany), then quality controlled using an Agilent 2100 Bioanalyzer (Agilent, Santa Clara, California, USA). The median RNA integrity number of samples used was 9.4
