Treatment protocol |
Mice were submitted to ischemia reprfusion protocol in vivo, IR was achieved through ligation of the left anterior descending (LAD) coronary artery for 45 min of ischemia, the heart was reperfused for 8h. HDL was injected at the onset of reperfuion. Sham operated mice did not have ischemia protocol.

Extraction protocol |
Total RNA was extracted using miRNeasy Mini kits according to manufacturer's protocol. Aftre reperfusion, the ventricles were collected and snap frozen for total RNA extraction.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 20 ug of cRNA were hybridized for 16 hr at 45C on GeneChip miRNA 4.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneChip Scanner 3000.

Data processing |
The data were analyzed with Partek Genomics Suite using RMA algorithm.
