Extraction protocol |
Total RNA was extracted from the liver tissues using TRIzol Reagent through chloroform/isopropanol purification. RNA quantity and purity was assessed using NanoDrop ND-1000. Pass criteria for absorbance ratios are established as A260/A280 ≥ 1.8 and A260/A230 ≥ 1.5. RIN values are assessed and examined using Agilent RNA 6000 Nano assay to determine RNA integrity.

Fluorescent aRNA targets were prepared from 1 μg total RNA samples using OneArray® Amino Allyl aRNA Amplification Kit (Phalanx Biotech Group, Taiwan) and Cy5 dye (GE Healthcare).

Hybridization protocol |
Total samples were hybridized using Phalanx OneArray Plus Protocol. Non-specific binding targets were washed away after 16 hours.

Scan protocol |
The slides scanned using Agilent 0.1 XDR Protocol.

Data processing |
GPR files were loaded into Rosetta Resolver® System (Rosetta Biosoftware) for data processing. The raw data normalized using the normalizeQuantiles function from 'limma' package (v. 3.30.7).
