Growth protocol Mice (strain C57/BL6) were kept in conventional M3 cages bedded with wood chips and paper nesting material in a temperature-controlled environment at 21±1°C, 45-55 % humidity on a 12-hr light/dark cycle. Food and water were provided ad libitum. Mice were allowed a 7-day acclimatisation period after arrival on site prior to experimental use. At 12 weeks of age, females were mated overnight and investigated for plug formation to indicate successful mating. Males were removed at this stage. In parallel, age-matched littermates were kept seperate. Plugged females were left for 72 hours after plug formation before sacrificing them using schedule 1 method. 2nd, 3rd, and 4th mammary glands were immediately removed and one flank used for fibroblast isolation. Fibroblasts were isolated as described by Ibrahim et al., 2017.
Extracted molecule total RNA
Extraction protocol Direct-zol™ RNA MicroPrep (Zymo Research) for freshly isolated fibroblasts (500-2,000 cells) as per manufacturer’s instructions and further described by Ibrahim et al., 2017. RNA amplification was subsequently performed using a NuGen-RNA amplification kit according to the manufacturer's protocol with the advised alterations for use in Illumina Genebead Microarray hybridisation.
