A novel method for quantitative sequencing enabling absolute quantification of template DNA molecule with sequence information was developed. For testing the concept of method, environmental samples from paddy field soil and beach sand were used.In addition, we validated the method with mock microbial communities which consist gDNA of Methanocaldococcus jannaschii, Halomonas elongata, Sulfolobus tokodaii, Bacillus subtilis subsp. subtilis, Streptomyces avermitilis, and Paracoccus denitrificans.
