miRNA array after p63 knock down in human squamous cell carcinoma

(Submitter supplied) A central challenge in human cancer therapy is the identification of pathways that control tumor cell survival and chemosensitivity in the absence of functional p53. The p53-related transcription factors p63 and p73 exhibit distinct, p53-independent roles in development and cancer: p73 promotes genome stability and mediates chemosensitivity, while p63 largely lacks these p53-like functions and instead promotes proliferation and cell survival. more...

Dysregulated deltaNp63alpha negatively regulates p73-mediated maspin expression

(Submitter supplied) The p53 homologue, p63, is critical for normal epidermal development. While overexpression of deltaNp63alpha has been reported in squamous cell cancers, the contribution of p63 to cancer pathogenesis remains unclear. We previously demonstrated that overexpressed deltaNp63alpha aberrantly maintains proliferation of primary epidermal keratinocytes under conditions that normally induce growth arrest and differentiation. more...

RNA-Seq analysis comparing p53-null versus ΔNp63Δ/Δ;p53-null or ΔNp73Δ/Δ;p53-null thymic lymphoma tumors

(Submitter supplied) We performed an RNA-Seq analysis comparing thymic lymphoma tissues from the p53-null(n=2) and ΔNp63Δ/Δ;p53-/- (n=3) or ΔNp73Δ/Δ;p53-/-(n=3). Mice at 10 weeks of age were injected with either Ad-mCherry or Ad-CRE-mCherry to delete ΔNp63/ΔNp73 in the thymic lmyphomas. We aimed to test by deleting the DNp63/DNp73 in these p53-deficient tumors will mediate tumor regression and analyze the expression profile of the genes

Identification of target genes regulated by ΔNp63α in SaOS-2 cells stably expressed with ΔNp63α

(Submitter supplied) To identify the target gene of ΔNp63α, in osteosarcoma cells, we performed the global gene expression profiling in SaOS- 2 cells stably overexpressing the ΔNp63α gene (SaOS- 2-ΔNp63α). ΔNp63α target genes were identified by comparison with genes expressed from SaOS-2-EV transfected with an empty vector ( SaOS-2-EV).

Expression data from H226 squamous cell carcinoma cells

(Submitter supplied) To define the contribution of p53 versus DNp63a to gene expression control, we performed a series of microarray assays for cells where p53 was activated using Nutlin-3 or DNp63a was knocked down using a stably transduced tetracycline inducible shRNA. We determined that p53 transcriptionally activates and DNp63a transcriptionally represses a non-overlapping subset of genes.
