Effect of obacunone on Salmonella Typhimurium transcriptome

(Submitter supplied) Obacunone is a limonoids present in Citrus species. We previously reported that obacunone was inhibitory to the cell-cell signaling in Vibrio harveyi and Escherichia coli O157:H7. In the present work we evaluated the effect of obacunone on the food borne pathogen Salmonella Typhimurium LT2 using cDNA microarray. The results demonstrate that obacunone exerts an antivirulence effect on S. Typhimurium LT2 by repressing SPI1 and SPI2. more...

Salmonella Typhimurium LT2: Control versus naringenin

(Submitter supplied) Salmonellosis is one of the leading health problems worldwide. With the rise of drug resistance strains it has become imperative to identify alternative strategies. Naringenin, a flavonone, is present predominantly in grapefruit. Previously we have demonstrated that naringenin is potent inhibitor of cell-cell signaling. In the present study, we provide evidence that naringenin specifically represses 24 genes in the Salmonella pathogenicity island 1, and down-regulates 17 genes involved in flagellar and motility; thereby, attenuating virulence and cell motility, respectively. more...

The nucleoid-associated protein HU controls three regulons that coordinate virulence and general physiology in Salmonella enterica serovar Typhimurium

(Submitter supplied) The regulon of genes controlled by the HU nucleoid-associated proteins was examined in Salmonella enterica serovar Typhimurium (S. Typhimurium). DNA microarrays were used to detect the effect on the bacterial transcriptome of knockout mutations in the hupA, the hupB and in both the hupA and hupB genes. Distinct but overlapping patterns of gene expression were detected in the transcriptome at each time point for each of the three mutants, pointing to the existence of not one but three regulons of genes controlled by the HU proteins. more...

Influence of Microbial and Host Cell Sublethal Heat Stress on S. Typhimurium Gene Expression

(Submitter supplied) Environmental stress contributes to the outcome of infection by impacting both microbial virulence and host susceptibility to infection. Thermal processing, commonly used for decontamination of poultry in the food industry, may elicit sublethal stress on resistant serovars of Salmonella. We employed traditional heat shock temperatures (42 and 48ºC), similar to avian body temperature and poultry processing conditions, to study gene expression of Salmonella enterica serovar Typhimurium. more...

Genome-wide screen for salmonella genes required for long-term systemic infection of the mouse.

(Submitter supplied) This datasets includes all arrays from the first passage through 129SvJ mice (28 days) and the second independent passage (29 days). The first passage arrays are the same as included in the experiment set "129SvJ Pass 1 28 dpi arrays" and the second passage arrays are the same as included in the experiment set "129SvJ Pass 2 28 dpi arrays". Input arrays are also included and collectively they represent two biological replicates for negative selection at ~4 weeks post infection. more...

Role of ProQ in Salmonella virulence - in vitro

(Submitter supplied) FinO-domain proteins such as ProQ of the model pathogen Salmonella enterica have emerged as a new class of major RNA-binding proteins in bacteria. ProQ has been shown to target hundreds of transcripts including mRNAs from many virulence regions but its role, if any, in bacterial pathogenesis has not been studied. Here, using a Dual RNA-seq approach to profile ProQ-dependent gene expression changes as Salmonella infects human cells, we reveal dysregulation of bacterial motility, chemotaxis and virulence genes which is accompanied by altered mitogen-activated protein kinase signaling in the host. more...

Expression analysis of Salmonella enterica serovar Typhimurium UK1 delta-iacP mutant

(Submitter supplied) Investigation of whole genome gene expression level changes in a Salmonella enterica serovar Typhimurium UK1 delta-iacP mutant, compared to the wild-type strain. IacP is resoponsible for the secretion of virulence effector proteins via the type III secretion system, thereby contributing the virulence of S. Typhimurium. The mutants analyzed in this study are further described in Kim et al. 2011. Role of Salmonella Pathogenicity Island 1 Protein IacP in Salmonella enterica Serovar Typhimurium Pathogenesis. more...
