Profiling of microRNA expression in lung cancer cells with up-regulation of Zeb1

(Submitter supplied) The Zeb1 transcriptional repressor plays a key role in metastasis through the down-regulation of genes that are strong inducers of epithelial differentiation and inhibitors of stem-ness. Here we report that Zeb1 controls the expression of numerous oncogenic and tumor suppressive microRNAs (miRs). Zeb1 stimulated pro-migratory cytoskeletal processes by down-regulating miR-34a and activated Rho GTPases through Arhgap1, a Cdc42 GTPase activating protein and novel miR-34a target gene. more...

Gene expression profiling of AC9 overexpression in 393P mice cells

(Submitter supplied) ADCY9 encodes AC9, which is 1 of 9 transmembrane AC isoforms (AC1-9) that catalyze the synthesis of cyclic AMP from ATP and are regulated primarily by heterotrimeric G-protein-coupled receptors. We ectopically expressed AC9 in 393P cells and found that, relative to empty vector controls, the AC9 transfectants had higher cyclic AMP levels and were more migratory and invasive in Boyden chambers. We examined the prognostic value of an AC9-regulated gene expression signature derived from 393P_AC9 cells and 393P_vector cells. more...

RNA-Seq analysis miR-455-3p overexpression effect on breast cancer cell line

(Submitter supplied) MiR-455-3p has been reported to suppress the expression of ROCK2, HOXB5, hTERT, and RUNX2, which are involved in multiple biological processes including renal fibrosis, chondrogenic development and differentiation, Alzheimer’s disease, preeclampsia, and tumorigenesis. Here, we showed that miR-455-3p broadly inhibited TGF-beta-induced EMT by repressing the expression of Smad2, ZEB1, and HDAC2 in breast cancer cells, and that miR-455-3p loss-of-function promotes the acquisition of cell invasive properties and the ability to metastasize. more...

The expression level of microRNA after knockdown of GATA3 in MCF-7 cells

(Submitter supplied) GATA3 is a basic and essential transcription factor that regulates many pathophysiological processes that is required for the development of mammary luminal epithelial cells. Loss-of-function GATA3 alterations in breast cancer are associated with poor prognosis. However, its regulation and function in miRNAs expression has been less explored.

miR-1199-5p and Zeb1: a novel double-negative feedback coordinating EMT and tumour cell invasion (mRNA-seq)

(Submitter supplied) We investigated the effect of miR-1199-5p, miR-200b-3p and miR-429-3p on gene expression profiles during TGFbeta-induced EMT in normal murine mammary gland cells by using the mRNA-sequencing. Our analysis demonstrates that miR-1199-5p and both miR-200 family members share only 6 target genes, indicating that besides regulating Zeb1 expression they exert distinct functions during EMT.

Transcriptional profiling of lung cancer cells transfected with Zeb1.

(Submitter supplied) To elucidate the mechanisms by which the mir-200 and the miR-183~96~182 cluster could regulate EMT and thus cellular migration, invasion and metastasis in NSCLC, we searched for common predicted targets of these microRNA families that might have a potential role in these biological processes. First we performed a cross comparison of multiple gene expression datasets from our mouse models of metastasis. more...

Identification of Metastasis-prone Lung Adenocarcinoma Cell Population That Is Sensitive to Notch Inhibition

(Submitter supplied) Tumor cells that give rise to metastatic disease are a primary cause of cancer-related death and have not been fully elucidated in patients with lung cancer. Here, we addressed this question by using tissues from a mouse that develops metastatic lung adenocarcinoma owing to expression of mutant K-ras and p53. We identified a metastasis-prone population of tumor cells that differed from those with low metastatic capacity on the basis of having sphere-forming capacity in Matrigel cultures, increased expression of CD133 and Notch ligands, and relatively low tumorigenicity in syngeneic mice. more...

Increased miR-708 expression in NSCLC and its association with poor survival in lung adenocarcinoma from Never Smokers

(Submitter supplied) We demonstrate that miR-708 is one of the most highly overexpressed miRNAs in non-small cell lung cancer. High level of miR-708 in tumor is also associated with a reduced overall survival in lung adenocarcinomas from never smokers. Functionally, miR-708 overexpression increases the proliferation, migration, and invasion in cultured cells and down regulates TMEM88, a negative regulator of Wnt signaling. more...
