miRNA expression profile from ANGPTL1 overexpression lung cancer cell

(Submitter supplied) ANGPTL1-overexpressed CL1-5 cells were used to identify miRNAs which regulate EMT, cancer invasion and metastasis. We used microRNA Taqman Low Density Array to identify candidate miRNAs

Profiling of microRNA expression in lung cancer cells with up-regulation of Zeb1

(Submitter supplied) The Zeb1 transcriptional repressor plays a key role in metastasis through the down-regulation of genes that are strong inducers of epithelial differentiation and inhibitors of stem-ness. Here we report that Zeb1 controls the expression of numerous oncogenic and tumor suppressive microRNAs (miRs). Zeb1 stimulated pro-migratory cytoskeletal processes by down-regulating miR-34a and activated Rho GTPases through Arhgap1, a Cdc42 GTPase activating protein and novel miR-34a target gene. more...

PRRX1 deficiency induces mesenchymal-epithelial transition through PITX2/miR-200-dependent SLUG/CTNNB1 regulation in hepatocellular carcinoma

(Submitter supplied) Metastasis is a major obstacle to better prognosis in patients with hepatocellular carcinoma (HCC). Mesenchymal-epithelial transition (MET) is the driving force for metastatic colonization in which E-cadherin reexpression is a critical procedure. It has been reported that the loss of paired-related homeobox transcription factor 1 (PRRX1) is required for cancer cell metastasis in vivo. However, the role of PRRX1 in MET and how its downregulation triggers E-cadherin reexpression are unknown. more...

Transcriptional profiling of lung cancer cells transfected with Zeb1.

(Submitter supplied) To elucidate the mechanisms by which the mir-200 and the miR-183~96~182 cluster could regulate EMT and thus cellular migration, invasion and metastasis in NSCLC, we searched for common predicted targets of these microRNA families that might have a potential role in these biological processes. First we performed a cross comparison of multiple gene expression datasets from our mouse models of metastasis. more...

Genes regulated by miR-203 in breast cancer cell line MDA-MB-231

(Submitter supplied) To investigate the mechanism through which miR-203 inhibited the breast cancer cell invasion, we overpression miR-203 in MDA-MB-231 cell line and performed a microarray to examine the genes which maybe targeted and down-regulated by miR-203.

The gene regulation of SPANXA in lung cancer

(Submitter supplied) SPANXA is identified as a cancer-testis antigens expressed in normal testis but dysregulated in various tumors. It plays unknown roles. This study sought to investigate the effects of the nuclear protein, SPANXA, on the biological behavior and global gene expression profiles of lung cancer cells. Expression microarray and bioinformatics analyses indicated that SPANXA mainly regulated genes involved EMT pathways in CL1-5 cells, a pattern that correlated with the altered behavior of CL1-5 cells observed after overexpressed SPANXA. more...

Angptl-7 knockdown RNA sequencing of 4T1 orthotopic transplants into rat mammary fat pads

(Submitter supplied) These samples are derived from tissues obtained from 4T1 spheroids transplanted orthotopically into the mammary fat pads of SRG OncoRats. 4T1 cell lines were established by viral transduction that stably express either a non-targeting control shRNA (NT4) or an shRNA targeting Angptl-7. 4T1 cells were cultured as spheroids in suspension for 24-hours prior to transplantation in 2% matrigel. 600,000 cells in a 1:1 mixture of matrigel and DMEM/F12 were transplanted orthotopically into the right T4 fat pads of SRG rats. more...
