Genomic pathways modulated by Twist in breast cancer

(Submitter supplied) Background: The basic helix-loop-helix transcription factor TWIST1 (Twist) is involved in embryonic cell lineage determination and mesodermal differentiation. There is evidence to indicate that Twist expression plays a role in breast tumor formation and metastasis, but the role of Twist in dysregulating pathways that drive the metastatic cascade is unclear. Importantly, the genes and pathways dysregulated by Twist in cell lines and mouse models have not been validated against data obtained from patient samples. more...

Gene expression profiling of FGFR1 KO mice

(Submitter supplied) A novel RCAS-Cre-IRES-PyMT (RCI-PyMT) virus was designed to specifically knockout genes of interest in tumors generated in appropriate mutant mouse hosts. We used this tumor knockout, or TuKO, strategy to concisely ablate fgfr1 in PyMT induced mammary tumors in K19-tva/fgfr1loxP/loxP mice. The similarly injected control K19-tva mice developed mammary tumors exhibiting high metastasis penetration to lung, making this an ideal model for breast cancer metastasis. more...

Exon-based clustering of tumor transcriptomes reveals alternative exons associated with metastasis in breast cancer

(Submitter supplied) Exonic profiling is an emerging topic in disease-associated transcriptome analysis, but is still facing major limitations. In this study, we performed a microarray-based analysis of the transcriptome at the exon level of mouse primary mammary tumors having different abilities to give rise to metastases. First, we developed a bioinformatics platform and used a stringent algorithm to identify 679 genes with differentially expressed exons in primary tumors of the 4T1 mouse model of mammary tumor progression. more...

Targeted Pten deletion plus p53-R270H mutation in mouse mammary epithelium induces aggressive claudin-low and basal-like breast cancer

(Submitter supplied) WAP-Cre:Ptenf/f:p53lox.stop.lox_R270H composite mice were generated by genetic crossing. In these mice, Pten is deleted and a R270H p53 mutation in the DNA binding domain is induced upon expression of Cre recombinase in pregnancy-identified alveolar progenitors. Tumors were characterized by histology, marker analysis, various bioinformatics methods, high-throughput (HTP) FDA-drug screen as well as orthotopic injection to quantify tumor initiating cells (TICs) and tail-vein injection to identify lung-metastasis. more...

Genome-wide gene expression profiling analysis of bone metastases of human non-small cell lung cancer (NSCLC) in mice

(Submitter supplied) Using a multiorgan metastasis model of human NSCLC cells (ACC-LC319/bone2) in NK cell-depleted SCID mice, we attempted to identify genes involved in the organ-selective nature of lung cancer-metastasis process, especially bone metastasis. Genome-wide gene expression profile of 15 metastatic lesions from three organs (bone, lung and liver) in this mouse model revealed lot of genes that seemed to reflect the organ specificity of metastatic cells. more...

Target genes associated to siRNA mediated repression of Pea3 and Erm transcription factors in mammary cancerous cells

(Submitter supplied) Pea3 and Erm are transcription factors belonging to the ETS family which are involved in tumorigenesis, particularly mammary oncogenesis. To elucidate the mechanism of Pea3/Erm induced tumorigenesis in mouse mammary cancerous MMT cells (ATCC CCL-51), we have down-regulated Pea3 or Erm expression in this cell line by using small interfering RNA and performed a microarray analysis using Applied Biosystems AB1700 technology to define target associated genes. more...

Genomic profiling of murine mammary tumors identifies potential personalized drug targets for p53-deficient mammary cancers

(Submitter supplied) Breast cancer is the second leading cause of cancer related death in American women. Patient care is complicated by inherent tumor heterogeneity that can be classified into at least six intrinsic subtypes. While targeted treatments are standard of care for most subtypes, there remains a clinical need for targeted therapies against basal-like tumors that are typically ‘triple negative breast cancers’. more...

The Rho Exchange Factors Vav2 and Vav3 Control a Lung Metastasis–Specific Transcriptional Program in Breast Cancer Cells

(Submitter supplied) The guanosine triphosphatases of the Rho and Rac subfamilies regulate protumorigenic pathways and are activated by guanine nucleotide exchange factors (Rho GEFs), which could be potential targets for anticancer therapies. We report that two Rho GEFs, Vav2 and Vav3, play synergistic roles in breast cancer by sustaining tumor growth, neoangiogenesis, and many of the steps involved in lung-specific metastasis. more...

A Splice Variant of HER2 Activates Key Signaling Cascades and Evokes Mammary Tumors and Metastases

(Submitter supplied) The Epidermal Growth Factor Receptor 2 (ERBB2 or HER2) is amplified and overexpressed in approximately 20% of invasive breast cancers and is associated with metastasis and poor prognosis. Here we describe the role of a constitutively active splice variant of HER2 (Delta-HER2) in human mammary epithelial cells. Overexpression of Delta-HER2 in human mammary cells decreased apoptosis and increased proliferation and expression of epithelial-to-mesenchymal markers. more...

A novel metastasis–associated lncRNA destabilizes p21 through cooperative interactions with the NF90/NF45 complex

(Submitter supplied) Long non-coding RNAs (lncRNA) have emerged as important regulators of gene expression at both the transcriptional and post-transcriptional levels. While altered expressions of lncRNAs have been observed in breast cancer (BCa) development, their roles in BCa progression and metastasis are still poorly understood. To identify novel BCa-associated lncRNA candidates, we have employed a high-density SNP array based approach to uncover lncRNA genes at intergenic region that are aberrantly expressed in BCa. more...
