Analysis of the molecular dialogue between gray mold (Botrytis cinerea) and grapevine (Vitis vinifera) reveals a clear shift in defense mechanisms during berry ripening

(Submitter supplied) Mature grapevine berries at the harvesting stage (MB) are very susceptible to the gray mold fungus Botrytis cinerea while veraison berries (VB) are not. We conducted simultaneous microscopic and transcriptomic analyses of the pathogen and the host to investigate the infectious process developed by B. cinerea on MB versus VB, and the plant defense mechanisms deployed to stop the fungus development. On the pathogen side, our genome-wide transcriptomic data revealed that B. more...

RNA-seq analysis of chitin-triggered transcriptional reprogramming in the Arabidopsis mapkkk5 mutant

(Submitter supplied) Perception of microbe-associated molecular patterns by host cell-surface pattern recognition receptors triggers a series of immune responses such as activation of mitogen-activated protein kinase (MAPK) cascades. In Arabidopsis thaliana, the receptor-like cytoplasmic kinase PBL27 interacts with the chitin receptor CERK1 and regulates chitin-induced MAPK activation. We found that the MAPK kinase kinase MAPKKK5 connects PBL27 to the downstream MAPK Kinases (MKK4/5) and MAPKs (MPK3/6). more...

Expression analysis of Arabidopsis peptidoglycan (PGN) recognition mutants treated with PGN

(Submitter supplied) Recognition of microbial patterns by host pattern recognition receptors is a key step in immune activation in multicellular eukaryotes. Peptidoglycans (PGN) are major components of bacterial cell walls and possess immunity-stimulating activities in metazoans and plants. Here, we show that PGN sensing in Arabidopsis thaliana requires three LysM domain proteins, LYM1, LYM3 and CERK1.

Chito-oligomer effect on seedlings

Analysis of 14-day-old Columbia-0 seedlings treated with hydrolyzed crab-shell chitin or chito-octamer. Chitin is a major component of fungal walls and insect exoskeletons. Results provide insight into the signaling pathways involved in activating defense genes in response to chito-oligomers.

Powdery Mildew- and Salicylic Acid-Induced Gene Expression in Grapevine

(Submitter supplied) Infection by the pathogen grape powdery mildew (Erysiphe necator) causes changes in the transcriptome of its susceptible host Vitis vinifera. Infection triggers the host to synthesize the signaling molecule salicylic acid (SA) which regulates the expression of a broad range of defense-related plant genes. In addition, it is hypothesized that E. necator directly modulates gene expression in V. vinifera via the haustorial complex. more...

Transcript profile of chitosan-treated Arabidopsis seedlings

(Submitter supplied) We treated Arabidopsis seedlings with chitosan and carried out a transcript profiling analysis (GeneChip microarrays) in order to identify genes and transcription factors regulated by chitosan. The results showed that jasmonate and defense responsive genes, camalexin and lignin biosynthetic genes were among genes up-regulated by chitosan. Several transcription factors are also strongly induced by chitosan. more...

Global transcriptional analysis suggests Lasiodiplodia theobromae pathogenicity factors involved in modulation of grapevine defensive response

(Submitter supplied) Purpose: Lasiodiplodia theobromae is a pathogenic fungus causing Botryosphaeria dieback in grapevine. Heat and drought stress was suggested to promote the disease incidence in recent years, therefore we decided to evaluate the effect of heat on in vitro experiment, adding grapevine wood to simulate the environment in the trunk during in field infection. Methods: Fungal mRNA was extracted from mycellium collected after 49 hours of growth without stress in the presence of GW (GW_noHS) or in its absence (VS_noHS). more...

The function of CEBiP in the chitin elicitor signaling in rice cells

(Submitter supplied) To see the function of CEBiP in the chitin elicitor signaling in rice cells, a plasmid for the gene-specific knock-down by RNA interference (RNAi) was constructed by using a sequence for the 3’-terminal region of CEBiP and transformed into the rice cells by using Rhizobium radiobacter (Agrobacterium tumefaciens). chitinoligomer was added (or not added) to the medium. For the control of them, rice cells that non transformed wild type was used. more...

O antigen acts as a shield to delay early plant immune recognition of the pathogenic bacterium, Xylella fastidiosa

(Submitter supplied) PAMP-triggered immunity (PTI) is the first line of plant defense against invading organisms. Initiated through the perception of conserved pathogen-associated molecular patterns (PAMPs), such as lipopolysaccharide or flagellin, PTI can provide early protection against a broad range of pathogens. Active suppression of PTI by microbial effector proteins, particularly those secreted by the Type III secretion system (T3SS), is a well-known strategy employed by bacterial plant pathogens that enables them to subvert PTI and successfully colonize their hosts. more...

Comparative transcriptomics of Central Asian Vitis vinifera accessions reveals distinct defense strategies against powdery mildew

(Submitter supplied) Grape powdery mildew (PM), caused by the biotrophic ascomycete Erysiphe necator, is a devastating fungal disease that affects most Vitis vinifera cultivars. We have previously identified a panel of V. vinifera accessions from Central Asia with partial resistance to PM that possess a Ren1-like local haplotype. In this study we show that in addition to the typical Ren1-associated late post-penetration resistance, these accessions display a range of different levels of disease development suggesting that alternative alleles or additional genes contribute to determining the outcome of the interaction with the pathogen. more...

A woody plant degrading pathogen effector targets host glucose signaling protein VvRHIP1 to promote virulence

(Submitter supplied) Lasiodiplodia theobromae is one of the causal agents of Grapevine trunk diseases, which becomes a tremendous threat to the grapevine production worldwide. Plant pathogens secrete diverse effectors to suppress host immune responses or regulate the host metabolism to promote diseases. Our results suggest that L. theobromae LtCre1 targets host VvRHIP1 to manipulate the sugar signaling pathway, through disrupting the association of VvRHIP1 and VvRGS1 complex.
