RNA-seq and ATAC-seq analysis of the role of PBRM1 under stress conditions

(Submitter supplied) We used RNA-seq to characterize the contributions of PBRM1 to gene expression in NmuMG cells with and without oxidative or low-glucose stress conditions. Further, we used ATAC-seq to characterize the effects of PBRM1 on chromatin accessibility in HK-2 and NMuMG cell lines. Our analyses reveal a role for PBRM1 in expression of genes associated with stress responses and with specific transcription factor families, such as the FOXO family.

The inactivation of PBRM1 is the second most frequent mutational event in kidney tumorigenesis. Here we show that in VHL-deficient ccRCC tumors, PBRM1 loss results in an altered PBAF complex that retains the association between SMARCA4 and ARID2 but disengages BRD7 from SMARCA4. PBAF loss leads to DNA damage induced inflammatory signalling through defective G2/M checkpoint maintenance

(Submitter supplied) PBRM1 deficiency in immortalised fibroblast cell line 1BR3-hTERT after ionising radiation treatment leads to more mitotic progression and more formation of micronucleated cells. Here, we performed total RNA-seq in 1BR3-hTERT parental line and PBRM1 knockout clones (PBRM1 KO3&KO5) under untreated (UT) or 48 hr post 6Gy irradiation condition.
