Anti-cancer agent sapphyrin PCI-2050 effect on lung cancer cell line: dose response

Analysis of A549 lung cancer cells following treatment with anti-cancer agent sapphyrin PCI-2050 or transcription inhibitor actinomycin D. Hydrophilic sapphyrins localize to tumors, generate oxidative stress, and inhibit gene expression.

Cultured A549 lung cancer cells treated with actinomycin D and sapphyrin PCI-2050

(Submitter supplied) The goal of this project was to characterize changes in gene expression in response to the anti-cancer agent sapphyrin PCI-2050. Cultured A549 human lung cancer cells were treated with sapphyrin PCI-2050 or actinomycin D, a known transcripitonal inhibitor. The gene expression profiles of drug-treated and control A549 cultures were determined using Human Genome U133 Plus 2.0 Arrays (Affymetrix, Santa Clara, CA). more...

Synthesis and Anticancer Properties of Water-Soluble Zinc Ionophores 2

(Submitter supplied) We have demonstrated that water-soluble zinc ionophores can be administered to mice at relatively high doses and inhibit the growth of A549 lung cancer cells grown in xenograft models. Gene expression profiles of tumor specimens harvested from mice four hours after treatment confirmed that the activation of stress responsive genes occurs in vivo. These findings lead us to propose that the pharmacologic delivery of zinc to tumors using water solubilized ionophores is a potential approach to cancer therapy. more...

Synthesis and Anticancer Properties of Water-Soluble Zinc Ionophores 1

(Submitter supplied) We have shown that water solubilized versions of a zinc ionophore increase intracellular concentrations of free zinc and have antiproliferative activity in exponential phase A549 lung cancer cultures. The gene expression profiles of A549 lung cancer cultures treated with the lead compound PCI-5002 reveal the activation of stress response pathways. Medium supplementation with zinc (25 μM) led to activation of additional oxidative stress response as well as apoptotic pathways. more...

Cytotoxic activity of HTI-286 in prostate cancer

(Submitter supplied) HTI-286, an analogue of hemiasterlin, interferes with microtubule dynamics and circumvents transport-based resistance to taxanes. In this study we evaluate the inhibitory effects of HTI-286 on human prostate cancer growth in vitro and in different in vivo models. The results show that HTI-286 is a potent inhibitor of proliferation and induced marked increases in apoptotic rates in all cell lines tested. more...

Hemiasterlin analog HTI-286 effect on docetaxel-resistant prostate cancer cell line

Analysis of prostate cancer LNCaP cells treated with the chemotherapeutic agent docetaxel or the hemiasterlin analog HTI-286. Like docetaxel, HTI-286 disrupts microtubule dynamics. But unlike docetaxel, HT-286 exhibits reduced multidrug resistance.

Profiling protease and protease inhibitor gene expression in tumors and their microenvironment

(Submitter supplied) To assist in the identification of proteases, their endogenous inhibitors, and proteins that interact with proteases or proteolytic pathways in biological tissues, a dual species oligonucleotide microarray has been developed in conjunction with Affymetrix, Inc. To investigate the performance of the array, gene expression profiles were analyzed in pure mouse and human samples and orthotopically implanted xenografts of human A549 lung and MDA-MB-231 breast carcinomas. more...

Motexafin Gadolinium and Zinc Induce Oxidative Stress Responses and Apoptosis in B-Cell Lymphoma Lines

(Submitter supplied) We report the effect of motexafin gadolinium (MGd) and exogenous zinc on intracellular levels of free zinc, oxidative stress, proliferation, cell cycle, and cell death in exponential phase human B-cell lymphoma and other hematologic cell lines. We find that increased levels of oxidative stress and intracellular free zinc precede and correlate with cell cycle arrest and apoptotic response. Treatment with exogenous zinc increased oxidative stress and, conversely, oxidative stress resulted in increased intracellular free zinc levels. more...

Microarray profiles of radiation response in the NCI60 cell lines

(Submitter supplied) The 60 cell lines of the NCI Anti-Cancer Drug Screen (NCI60) constitute the most extensively characterized in vitro cancer cell model, and have been tested for sensitivity to over 100,000 potential chemotherapy agents. We have used the NCI60 cell lines and three additional lymphoblast lines to develop a database of responses of cancer cells to ionizing radiation. We compared clonogenic survival, apoptosis induction, and gene expression response by microarray analysis. more...
