Onconase effect on mesothelioma cells in vitro

Analysis of malignant mesothelioma (MM) cell lines (M25, M29, and M49) exposed to Onconase for 24 hrs. Onconase, an RNA damaging therapeutic agent, induces apoptosis in MM cells. Results provide insight into the molecular mechanisms underlying Onconase action in MM.

Onconase Responsive Genes in Human Mesothelioma Cells: Implications for an RNA Damaging Therapeutic Agent

(Submitter supplied) Onconase represents a new class of RNA-damaging drugs. Mechanistically, Onconase is thought to internalize, where it degrades intracellular RNAs such as tRNA and double-stranded RNA, and thereby suppresses protein synthesis. However, there may be additional or alternative mechanism(s) of action. Microarray analysis was used to compare gene expression profiles in untreated human malignant mesothelioma (MM) cell lines and cells exposed to 5 ug/ml Onconase for 24 h.

Transcriptional profiling of human ovarian cancer cells treated with ribonucleases

(Submitter supplied) PE5 is a nuclear-directed human ribonuclease variant that exhibits selective cytotoxicity against cancer cells. Onconase is a natural cytotoxic ribonuclease that has reached advanced clinical trials for the treatment of different types of cancer. Onconase also posses established antiviral effects. In the present work, we have used global gene expression microarrays to study the effects caused by PE5 and onconase, and therefore elucidate the mechanism by which these ribonucleases induce cytotoxicity. more...

Gene expression profiling of diffuse malignant peritoneal mesothelioma

(Submitter supplied) Diffuse malignant peritoneal mesothelioma (DMPM) is a rapidly lethal malignancy. The comprehension of the molecular features of DMPM is of utmost importance for the fruitful management of the disease, especially in patients who fail standard treatments and have a poor prognosis due to the lack of effective alternative therapeutic options.

Mesothelioma differential expression

(Submitter supplied) To investigate the molecular events controlling malignant transformation of human pleural cells, we compared constitutive gene expression of mesothelioma cells to that of pleural cells. Using cDNA microarray and high-density filter array, we assessed expression levels of > 6500 genes. Most of the highly expressed transcripts were common to both cell lines and included genes associated with stress response and DNA repair, outcomes consistent with the radio- and chemo-resistance of mesothelioma. more...
