Psoriasis non-lesional skin response to interferon-γ

Analysis of non-lesional (NL) skin of psoriasis patients 24 hrs after a single intradermal injection of interferon-γ (IFN-γ) to the NL skin. Results suggest that IFN-γ may prime an inflammatory environment in the skin that shares many features with psoriasis.

A single intradermal injection of IFN-γ induces a psoriasis-like state in both non-lesional psoriatic and healthy skin

As IFN-γ is involved in many cellular processes, including activation of T cells and dendritic cells (DCs), antigen processing and presentation, cell adhesion and trafficking, and cytokine and chemokine production, IFN-γ-producing Th1 cells were proposed to be integral to the pathogenesis of psoriasis. Type I Interferon: Potential Therapeutic Target for Psoriasis?

(Submitter supplied) We observed robust overexpression of type I interferon (IFN)–inducible genes and genomic signatures that indicate T cell and dendritic cell infiltration in lesional skin. Post-therapeutic relapse of psoriasis associated with CD11a blockade is associated with T cells and inflammatory myeloid DCs

(Submitter supplied) To understand the development of new psoriasis lesions, we studied a group of moderate-to-severe psoriasis patients who experienced a relapse after ceasing efalizumab (anti-CD11a, Raptiva, Genentech). There were increased CD3+ T cells, neutrophils, CD11c+ and CD83+ myeloid DCs, but no increase in CD1c+ resident myeloid DCs. Single cell analysis of CD1a-restricted Group A streptococcus (GAS)-reactive peripheral and cutaneous T cells

MicroRNA profiling of psoriatic skin identifies 11 miRNAs associated with disease severity

(Submitter supplied) MicroRNAs (miRNAs) are small non-coding RNAs that have emerged as central regulators of gene expression and powerful biomarkers of disease. Much is yet unknown about their role in psoriasis pathology. Spatial transcriptomics stratifies psoriatic disease by emergent cellular ecosystems

(Submitter supplied) The skin is recognized as a window into the immunopathogenic mechanisms driving the vast phenotypic spectrum of psoriatic disease. To better decipher the cellular landscape of both healthy and psoriatic skin, we employed spatial transcriptomics (ST), a ground-breaking technology that precisely maps gene expression from histologically-intact tissue sections. Findings gleaned from computationally integrating our 23 matched lesional and non-lesional psoriatic and 7 healthy control samples with publicly-available single cell RNA seq data sets provided insights into the emergent cellular ecosystems at play in psoriatic disease.
