Control and fatty acid-loaded macrophages (2 x 10e6/well) were treated with LPS (10 ng/ml) or vehicle for 24 hrs prior to extraction of the RNA as described above.

Murine peritoneal macrophages, harvested 4 or 5 days after injecting 1.0 ml 3% Brewer’s thioglycollate (Beckton Dickenson/BBL) i.p., were allowed to adhere to plastic wells for 4-6 hours in complete RPMI (52). The cells were then washed twice with PBS and cultured further in DMEM without phenol red (Sigma D5030) that contained 0.5% low endotoxin fetal bovine serum (Hyclone), 1 g/L (5.5 mM) glucose, 44 mM NaHCO3, 1 mM sodium pyruvate, 2 mM L-glutamine, 100 U/ml penicillin G, and 100 µg/ml streptomycin (“cDMEM”).

RNA was extracted with Trizol, followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the manufacturer’s protocol. Quality control was performed using the Agilent Bioanalyser.
