Treatment protocol |
Mice were injected with LPS (from Escherichia coli O26:B6; 0.1 mg/kg) or saline solution, and sacrificed 6 hrs later.

Growth protocol |
4 weeks old mice were maintained an iron replete (Tmprss6 KO mice) or an iron deficient (control littermates) diet for 3 weeks.

Extraction protocol |
Total liver RNA was extracted with Trizol reagent, followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyser.

Total RNA was first reverse transcribed into cDNA and then in vitro trascribed into biotin-UTP labelled cRNA using the Illumina TotalPrep RNA Amplification Kit (Ambion, Austin, TX)

Hybridization protocol |
750 ng of cRNA in 11 µl (150 ng/µl) were hybridized to the BeadChip Array at 58°C overnight. The fluorescent signal was developed through a streptavidin-Cy3 staining step.

Scan protocol |
Beadchips were imaged using the Illumina BeadArray Reader; the software Illumina® GenomeStudio was used to take fluorescent hybridization signals.
