Sample GSM1164071 Query DataSets for GSM1164071
Status Public on Jan 25, 2016
Title Olfactory_Chlorpyrifos/Malathion_High_8
Sample type RNA

Source name Chlorpyrifos/Malathion High
Organism Oncorhynchus kisutch
Characteristics tissue: olfactory rosettes
developmental stage: juvenile
body weight: 15 +/- 5.7g
age: 1 year
Biomaterial provider The University of Washington (Seattle, WA).
Treatment protocol Pesticide stock solutions or carrier solutions containing no pesticide were added to 70L hatchery water in 90L glass aquaria containing eight individual fish. The aquaria were contained within a large chilled recirculating water bath (11C) and received aeration. Fish were exposed for 24h.
Extracted molecule total RNA
Extraction protocol The olfactory rosettes were dissected for subsequent RNA isolation. Total RNA was isolated using the RNeasy Mini Kit (Qiagen Inc, Valencia, CA) according to the vendor's protocol and stored at -80C.
Label Cy5 fluorescent label
Label protocol For microarray hybridization, RNA was processed for hybridization to the Agilent-025055 Koop Lab Salmonid 44K Array through a core facility in the Functional Genomics Laboratory in the Center for Ecogenetics and Environmental Health at the University of Washington according to the manufacturer's instructions.

Hybridization protocol The standard hybridization protocol was used as recommended by Agilent
Scan protocol Arrays were scanned using an Agilent DNA microarray scanner through the University of Washington Functional Genomics Laboratory in the Center for Ecogenetics and Environmental Health (http://depts.washington.edu/ceeh/ServiceCores/FC1/FC1.html).
Data processing Expression values were first background-corrected by subtracting the median background intensity, and then normalized using variance stabilization normalization (VSN, Huber et al, doi 10.1093/bioinformatics/18.suppl_1.S96).
