Title |
Olfactory_Chlorpyrifos_Medium_2 |
Sample type |
RNA |
Source name |
Chlorpyrifos Medium
Organism |
Oncorhynchus kisutch |
Characteristics |
tissue: olfactory rosettes developmental stage: juvenile body weight: 15 +/- 5.7g age: 1 year
Biomaterial provider |
The University of Washington (Seattle, WA).
Treatment protocol |
Pesticide stock solutions or carrier solutions containing no pesticide were added to 70L hatchery water in 90L glass aquaria containing eight individual fish. The aquaria were contained within a large chilled recirculating water bath (11C) and received aeration. Fish were exposed for 24h.
Extracted molecule |
total RNA |
Extraction protocol |
The olfactory rosettes were dissected for subsequent RNA isolation. Total RNA was isolated using the RNeasy Mini Kit (Qiagen Inc, Valencia, CA) according to the vendor's protocol and stored at -80C.
Label |
Cy5 fluorescent label
Label protocol |
For microarray hybridization, RNA was processed for hybridization to the Agilent-025055 Koop Lab Salmonid 44K Array through a core facility in the Functional Genomics Laboratory in the Center for Ecogenetics and Environmental Health at the University of Washington according to the manufacturer's instructions.
Hybridization protocol |
The standard hybridization protocol was used as recommended by Agilent
Scan protocol |
Arrays were scanned using an Agilent DNA microarray scanner through the University of Washington Functional Genomics Laboratory in the Center for Ecogenetics and Environmental Health (http://depts.washington.edu/ceeh/ServiceCores/FC1/FC1.html).
Data processing |
Expression values were first background-corrected by subtracting the median background intensity, and then normalized using variance stabilization normalization (VSN, Huber et al, doi 10.1093/bioinformatics/18.suppl_1.S96).
