Treatment protocol |
[collection protocol] A whole-stream early-morning urine specimen was collected into sterile tubes before renal biopsy and processed within 3 hours. Urine sample was centrifuged at 3,000 g for 30 min and at 13,000 g for 5 min at 4 °C. Urinary cell pellet was lysed by TRIzol® Reagent (Invitrogen™). Specimens were then stored at –80 °C until use.

About 1ug total RNA were polyA tailed and labled with FlashTag™ Biotin HSR Labeling Kit. This kit can be used to label any RNA sample, including total RNA, severely degraded RNA, plant RNA, and low molecular weight RNA.

The labled RNA were hybridized for 16 hr at 48C on Affymetrix miRNA Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneChip® Scanner 3000.

The data were analyzed with miRNA QCTool using Affymetrix default analysis settings and quantile as normalization method.
