Treatment protocol 5XFAD mice were treated by i.p. injection of human peptide amylin from AnaSpec (Fremont, CA, USA) or 1x phosphate-buffered saline (PBS) solution , while WT mice were treated i.p. with 1x PBS solution, on daily basis for 10 weeks.
Growth protocol For this study, all mice were maintained in microisolator cages in the Animal Facilities of Boston University School of Medicine, Boston, MA, USA. All animal procedures were in accordance with the National Institutes of Health Guide for the Care and Use of Laboratory Animals and were approved by the Boston University Animal Care and Use Committee.
Extracted molecule total RNA
Extraction protocol Total RNAs from brain tissues were extracted using QIAzol lysis reagents according to the manufacturer's instructions.
Label biotin
Label protocol Biotin labeling was performed using the Ambion WT Expression Kit (Life Technologies, Grand Island, NY) according to the manufacturer's protocol, followed by the GeneChip WT Terminal Labeling and Controls Kit (Affymetrix, Santa Clara, CA).
 
Hybridization protocol The labeled, fragmented cRNA was hybridized to the GeneChip Mouse 1.0 ST Array for 18 hours in a GeneChip Hybridization oven 640 at 45oC with rotation (60 rpm).  The hybridized samples were washed and stained using an Affymetrix fluidics station 450.
Scan protocol After staining, microarrays were immediately scanned using an Affymetrix GeneArray Scanner 3000 7G Plus.

Data processing Raw Affymetrix CEL files were normalized to produce probeset-level expression values using the implementation of the Robust Multiarray Average (RMA) in the oligo Bioconductor R package.
