Growth protocol |
The human induced pluripotent stem cells were cultured on SNL-feeder cells with Primate ESC medium (ReproCELL) supplemented with basic FGF and Activin A at 37 ÂșC in a humidified incubator with 5% CO2. In Samples 1-4, the cells were cultured with different concentrations of basic FGF (0, 4, 20, and 100 ng/ml) and without Activin A. In Samples 5-8, the cells were cultured with 4 ng/ml basic FGF and with different concentrations of Activin A (0, 4, 20, and 100 ng/ml).

Extraction protocol |
RNA was prepared using the miRNeasy Purification kit (Qiagen) following the manufacturer's recommendations. RNA was quantified using a NanoDrop-1000 spectrophotometer and quality was monitored with the Agilent 2100 Bioanalyzer (Agilent Technologies).
