Extraction protocol |
Frozen heart tissues were grounded into powder under liquid nitrogen and lysed by TRIZOL reagent (Invitrogen, Life Technologies Inc., Frederick, MA, USA). Total RNA + miRNAs was extracted according to manufacturer’s protocol, followed by treatment with Dnase I (Rnase-free 2U/µL, Life Technologies Inc., Carlsbad, CA, US).

The microarray chips were then washed and scanned using InnoScan700, microarray scanner (Innopsys, Carbonne, France) and analysed on Mapix® Ver4.5 software (Innopsys, Carbonne, France).

Data processing |
Background-subtracted median signal intensities were normalized using software GeneSpring gx v.13 with quantile normalization. Fold change analysis was performed using the same software. Unpaired ttest was performed using the raw signals to eliminate those miRNAs that were not significant between 2 groups. miRNAs with raw signal intensity >300 at least in 1 group were further selected. The final differentially expressed miRNAs were selected based an absolute fold change ≥ 1.2 and ≤ -1.2.
