Sample GSM2160035 Query DataSets for GSM2160035
Status Public on Sep 05, 2017
Title MI Day2
Sample type RNA

Source name rat heart tissue
Organism Rattus norvegicus
Characteristics strain: Wistar
Sex: male
treament: Rat undergone lateral thoracotomy and pericardiactomy and LAD ligation
Treatment protocol Rats in Sham Day 2 group underwent lateral thoracotomy and percardiotomy. Hearts were harvested 2 days post surgery
Extracted molecule total RNA
Extraction protocol Frozen heart tissues were grounded into powder under liquid nitrogen and lysed by TRIZOL reagent (Invitrogen, Life Technologies Inc., Frederick, MA, USA). Total RNA + miRNAs was extracted according to manufacturer’s protocol, followed by treatment with Dnase I (Rnase-free 2U/µL, Life Technologies Inc., Carlsbad, CA, US).
Label Hy 3
Label protocol Total RNA (1 µg for Day 2 and 500ng for D14) was 3′-end–labeled with Hy3 dye using the miRCURY LNATM Power Labeling Kit (Exiqon, Denmark)

Hybridization protocol Samples were then hybridized on miRCURY LNATM Arrays using MAUI® hybridization system
Scan protocol The microarray chips were then washed and scanned using InnoScan700, microarray scanner (Innopsys, Carbonne, France) and analysed on Mapix® Ver4.5 software (Innopsys, Carbonne, France).
Description Total RNA (1 µg) was 3′-end–labeled with Hy3 dye and hybridized on miRCURY LNA™ Arrays
Data processing Background-subtracted median signal intensities were normalized using software GeneSpring gx v.13 with quantile normalization. Fold change analysis was performed using the same software. Unpaired ttest was performed using the raw signals to eliminate those miRNAs that were not significant between 2 groups. miRNAs with raw signal intensity >300 at least in 1 group were further selected. The final differentially expressed miRNAs were selected based an absolute fold change ≥ 1.2 and ≤ -1.2.
