Monocytes (CD14+ cells) were isolated with Tuek4 Ab (Dako M0825) and IgG2a+b magnetic beads (Miltenyi) by AutoMacs. They were cultured with CM from each of the 2 stromal cell lines

| Growth protocol |
Cells were cultured for 2 days in RPMI1640 medium supplemented with 10% FBS/2mM glutamine at 37ºC in 5% CO2. Conditioned medium from the stromal cell lines was collected from 3 million stromal cells grown for 5 days. It was clarified by centrifugation and diluted 1:1 with fresh supplemented RPMI1640 medium as described above just prior to use.

| Data processing |
Affymetrix CEL files were imported into GeneSpring 7.3.1 and samples were preprocessed with GC-RMA to obtain the normalized natural hybridization values. The Per gene normalization feature of Genespring, in which each measurement is divided by the median of its measurements in all simples, was also applied.
