Growth protocol Fetal and adult erythroblasts were generated from CD34+ hematopoietic stem cells using a two-phase serum-free culture as described previously (Sankaran VG et al. Science 2008).

Extraction protocol We extracted genomic DNA with the Gentra Puregene Cell Kit (Qiagen)
Label C-Bio and A-DNP
Label protocol 4 μL of genomics DNA at 50ng/μL (approximately 200ng of genomic DNA) was whole genome amplified and fragmented.
 
Hybridization protocol Resuspended DNA was hybridized to locus-specific probes bound to the surface of each BeadChip. Unhybridized or non-specifically hybridized DNA was removed by gentle washing of BeadChips. DNA that remained specifically bound to the BeadChip was then genotyped using a fluorescently labeled single base extension assay
Scan protocol Each BeadChip was scanned and analyzed using the Illumina iScan Reader.

Genomic DNA extracted from erythroblasts was genotyped using Infinium HumanOmniExpress-12 v1.1 Genotyping BeadChips (Illumina).
Data processing Genomic DNA extracted from erythroblasts was genotyped using Infinium nfinium HumanOmniExpress-12 v1.1 Genotyping BeadChips (Illumina). Scanned images were analyzed using Illumina’s GenomeStudio2011.1, with the following parameters: A) cluster algorithm GenTrain 2.0, B) No-Call threshold of 0.15 checked. The data analysis process was performed using the standard Illumina HumanOmniExpress-12v1-1_B cluster file.
Processed data include: Genotype: A,B,or - (no call), Theta, R, B Allele frequency and Log R Ratio.
