Treatment protocol |
Mice were randomized based on tumor size and allocated to treatment groups and then treated with vehicle (0.5% carboxylmethylcellulose, CMC, p.o.) or RAD140 100 mg/kg. Treatment lasted 51 days before mice were euthanized and tumor specimen were snapfrozen.

HBCx-22 PDX model were established by implanting tumor fragments subcutaneously in the flank female athymic nude mice (Foxn1nu).

Extraction protocol |
Total RNA from xenograft samples and cells was extracted using the RNeasy Mini Kit (Qiagen), with a DNase incubation step included to ensure complete removal of genomic DNA. Purified RNA samples were converted into cDNA libraries using the TrueSeq Stranded mRNA sample preparation kit (Illumina, San Diego, CA). Libraries were quantified, normalized, and pooled before being subjected to HiSeq 2×50bp paired end sequencing on an Illumina sequencing platform.
