Treatment protocol |
Using tungsten needle and forceps, FGF-2 soaked beads were implanted in the posteror mesoderm of HH23 Chicken limb buds about 500microns from the distal tip. Embryos were incubated for an additional 3hrs then treated tissue was harvested and flash frozen using liquid nitrogen.

Growth protocol |
Fertile chicken eggs were incubated in a humidified chamber for 4 days or until embryos reached desired developmental stage (HH23).

RNA was extracted using QIAGEN RNeasy Plus Micro kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyser. RNA libraries were prepared for sequencing using standard Illumina protocols
