For every sample, transgenic hearts were dissected, the GFP-positive tissues pooled, dissociated to obtain single cells, and purified using FACS. 5000 cells per sample were gated and used for RNA isolation (Macherey-Nagel, nucleospin II). RNA was subjected to 2 rounds of linear amplification and biotin labeling (Ambion). Within each group 6 labeled antisense RNA samples were hybridized to separate Illumina Mouse-Ref-6 v1.1 BeadChips.
