Data processing |
Illumina Casava1.7 software used for basecalling. The clean reads were obtained by eliminating the low-quality reads, empty adapters, reads shorter than 18 nt. Sequences of tRNA, rRNA, snRNA, snoRNA and piRNA were removed by similarity search using the Rfam12.1 and piRNABank database. miRNAs were identified by blasting the rest sequences with miRbase (V21.0) and referring to the locust genome Then the RPKM value were calculated for expression level quantification.
