Treatment protocol |
tumor implant protocol: Mice were implanted intradermally with 5x10^5 B16 tumor cells on Day 0. Mice were injected intratumorally with MPL and CD40 and injected intraperitoneally with PD1 between Day 7-9. Other mice were injected intraperitoneally with PD1 alone. Treatment was given 2x/week and then sacrificed 1 week after start of trx. organ harvest protocol: Mice were sacrificed and the distant tumors were dissected and mechanically dissociated into single cell suspensions.

Biotinylated sense-strand DNA targets are generated from 50ng of total RNA following standard protocol using the WT Plus Reagent kit (Applied Biosystems GeneChip WT Plus Reagent Kit Target Preparation Manual).

Hybridization protocol |
Following fragmentation and labeling, 3.5ug of fragmented and labeled ss-cDNA was hybridized for 16hr at 45C on Clariom S Mouse HT array plates on the GeneTitan MultiChannel Instrument.

Scan protocol |
After hybridization was completed the washing, staining and scanning of the arrays was completed on the GeneTitan MultiChannel Instrument.
