Status Public on Jul 30, 2020
Title SPF45 siRNA treated cells #2
Sample type SRA

Source name HEK293 cells
Organism Homo sapiens
Characteristics cell line: HEK293
genotype/variation: SPF45 siRNA
Treatment protocol Six independent total RNAs derived from HEK293 cells treated with three control siRNAs
Growth protocol HEK293 cells were cultured in Dulbecco’s modified Eagle’s medium (DMEM; Wako) supplemented with 10% fetal bovine serum (FBS).
Extracted molecule total RNA
Extraction protocol Total RNAs were prepared with NucleoSpin RNA kit (Macherey-Nagel).
rRNA depletion was performed by RiboMinus Eukaryote System v2 (Invitrogen–Thermo Fisher Scientific). The preparation of RNA libraries were performed using NEBNext Ultra RNA Library Prep Kit for Illumina (New England Biolabs).

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina HiSeq 2500

Data processing RNA-seq reads were aligned to the hg19 genome assembly using Tophat2
bigwig files were generated using bam2wig then followed by wigToBigwig
Genome_build: hg19
Supplementary_files_format_and_content: bigwig
