Treatment protocol For Hypoxia_Normoxia_5days experiment cells were either placed under hypoxia (1% oxygen) or normoxia (18.6% oxygen) for 5 days. For DMOG experiment, cells were treated either with carrier control (EtOH) or with 3mM dimethyloxalylglycine (DMOG) for 24 hours.
Growth protocol Human GBM cell line A172 was obtained from ATCC. Cells were cultivated in complete DMEM medium. At 80-90% confluency cells were harvested and seeded for experiments. 24 hours after seeding, a medium change was effected and treatments were carried out.
Extracted molecule total RNA
Extraction protocol Total RNA was harvested from cultured cells using the RNeasy Mini Kit (Qiagen, 80204)

Label protocol The Affymetrix WT PLUS Reagent Kit is used to generate labeled ss-cDNA from input amounts of 100ng total RNA
 
Hybridization protocol According to Affymetrix WT PLUS Reagent Kit user manual, 5.5 ug of fragmented and labeled ss-cDNA were hybridized for 17 hr at 45°C on Affymetrix Human Gene 2.0 ST Arrays.
Scan protocol Gene Expression Microarrays were scanned using the Affymetrix GeneChip® Scanner 3000 according to GeneChip® Expression Wash, Stain and Scan Manual for Cartridge Arrays (P/N 702731)

Data processing The Affymetrix microarray chips “human gene 2.0 ST” were analyzed using the oligo package and annotated using NetAffx4 OtCEL files were imported from disk or ded by RMA normalization and summary.
Processed data are presented as log2 normalized RMA values. The Hypoxia_Normoxia_5days and DMOG experiments were analyzed separately and included in tabs in the file Hypoxia_DMOG_normalized.xlsx.
