HMB supplementation began one week prior to tumor cell injection, with mice randomized to receive either Ctrl (AIN-93G) or Ctrl+HMB, once tumors were established following injection of 200K PANC02 cells s.c. mice were treated with 200µg anti-PD1 every 3 days.

GeneChip® WT PLUS Reagent Kit (Affymetrix) was used to generate sense-strand cDNA from total RNA. Following synthesis of sense-strand cDNA, the cDNA was fragmented and labeled with the kit. Fragmented and labeled cDNA was used to prepare a hybridization cocktail with the Affymetrix GeneTitan Hybridization Wash and Stain Kit for WT Arrays
