Treatment protocol MCF7 cells are seeded on culture plate 2 days prior of experiment. On the day of the experiment, for the fast group, the cells were changed to fresh DMEM complete medium for 4h before treatment to ensure the viability. Then the cells were changed to glucose-free DMEM medium for 4 hours before collection. For the refed group, cells were also changed to fresh DMEM medium for 4h and followed by glucose-free DMEM medium for 4 hours. Then the cells were refed by changing to complete DMEM before collection.
Growth protocol MCF7 cells were maintained in DMEM supplemented with 10% fetal bovine serum (FBS) and antibiotics in humidified atmosphere with 5% CO2 at 37C.
Extracted molecule total RNA
Extraction protocol RNA was harvested using Invitrogen TRIzol according to manufacturer's instruction.
1.0 ug of total RNA was used for the construction of sequencing libraries. Sequencing libraries were generated using NEBNext® UltraTM RNA Library Prep Kit for Illumina® (NEB, USA).
