Growth protocol |
Cultured in RPMI 1640 medium (Life Technologies, Inc., Grand Island, NY) containing 10% heat-inactivated fetal bovine serum (FBS; BioWhittaker, Verviers, Belgium) at 37°C in a humidified atmosphere of 5% CO2 and passaged every 3 days.

Total RNA was prepared using QIAGEN RNeasy mini kit (QIAGEN, Inc., Valencia, CA). The quality of RNA was checked using Agilent Technologies 2100 Bioanalyzer (Agilent, Palo Alto, CA).

Biotin-labeled cRNA was synthesized from 1-µg total RNA using CodeLink Expressioin Assay Reagent Kit (Amersham) and Biotin-11-UTP (PerkinElmer) according to the manufacturers’ protocols.

Hybridization protocol |
Each cRNA sample was fragmented, and hybridized with a CodeLink UniSet Human 20K I Bioarray™ using Parallel Processing Kit™ (CodeLink) for 18 h at 37°C in an Innova™ 4080 incubator (New Brunswick Scientific) shaking at 300 rpm. The hybridized arrays were rinsed with 0.75 x TNT buffer at 46°C for 1 h, labeled with Streptavidin-Cy5 at room temperature for 30 min, rinsed with TNT buffer and 0.05% Tween 20, and dried by centrifugation.

The arrays were scanned using Agilent DNA Microarray Scanner™ (Agilent, PaloAlto, CA, USA).

Expression profiles of exponentially growing ovarian cancer cell lines were compared with their drug sensitivity.
