Extraction protocol |
Total RNA was isolated in triplicates from myoblasts with Pax7 overexpression, Pax3 overexpression and empty vector control using RNeasy mini kit (Qiagen). The purity of RNA was analyzed by Bioanalyzer (Agilent Technologies, Santa Clara CA). All samples showed RNI > 9.0 and were used for subsequent labeling and hybridization.

Transcript cluster identifiers (TCID) of the Affymetrix MoGene 1.0 chipset were RMA normalized using the xps Bioconductor package in R-2.10.0 with the Affymetrix provided MoGene-1_0-st-v1.r4 chip layout and scheme files. The CHTAP chips were used as controls and Pax7, Pax3 overexpression as the treatment.
